Dog HEPATOPAC® co-cultures are derived from highly characterized donors. Plates may be customized to be a single donor, pooled donor or multiple donors in different wells on the same plate.

Dog HEPATOPAC kits are shipped with all necessary media and an instructions for use manual. Dosing can begin two days after receiving the kit.

Validation

Dog HEPATOPAC co-cultures remain viable and highly functional for four weeks. The co-cultures secrete albumin, synthesize urea, display functional bile  canaliculi, and metabolize compounds using active Phase I and Phase II drug metabolism  enzymes. The following data illustrates the morphology, liver function, and metabolizing enzyme activity expected from dog HEPATOPAC co-cultures.

Morphology

Dog HEPATOPAC co-cultures were created and maintained in vitro for over four weeks. Over this time there were no substantial  changes to the morphology of micropatterned co-cultures and the polygonal shape was maintained. The pictures below illustrate the micropatterned HEPATOPAC islands and the interface of hepatocytes and stromal cells at nine days in culture.

Liver Function

As shown in the chart below, Promega’s P450-Glo CYP450 Assay was used to assess CYP activity over a 20-day period in a dog HEPATOPAC co-culture. The data illustrates the long-term viability of the HEPATOPAC co-culture.
